چکیده

Despite the considerable progress in both the diagnosis and treatment of breast cancer, the disease remains the second leading cause of cancer-related deaths among women. Drug therapies directed at the estrogen receptor, the progesterone receptor, and human epidermal growth factor 2 receptor (HER2/neu) have resulted in improved clinical outcomes in some breast cancers. However, it is still a formidable challenge to treat those tumors that do not express the aforementioned molecular targets (Sorlie et al., 2001; Sørlie et al., 2003; Schneider et al., 2008). Fangchinoline (Fan) is a bis-benzylisoquinoline alkaloid, extracted from the Chinese medicinal herb Radix Stephania tetrandrae, dry roots of Stephaniae tetrandrine S. Moore (Menispermaceae). Fan shares structural features with tetrandrine, another functional compound extracted from Radix Stephania tetrandrae. Tetrandrine has been shown to be a potent calcium channel blocker and demonstrates anti-tumour activity in various cancer cells (King et al., 1988). Hsu et al reported that tetrandrine

Long non-coding RNA FOXO1 inhibits lung cancer cell growth through down-regulating PI3K/AKT signaling pathway

Objective(s): Lung cancer is one of the most common malignant tumors, which seriously threatens the health and life of the people. Recently, a novel long non-coding RNA (lncRNA) termed lncFOXO1 was found and investigated in breast cancer. However, the effect of lncFOXO1 on lung cancer is still ambiguous. The current study aimed to uncover the functions of lncFOXO1 in l...

Akt/protein kinase b and glycogen synthase kinase-3beta signaling pathway regulates cell migration through the NFAT1 transcription factor.

The phosphoinositide 3-kinase (PI3K) pathway regulates a multitude of cellular processes. Deregulation of PI3K signaling is often observed in human cancers. A major effector of PI3K is Akt/protein kinase B (PKB). Recent studies have pointed to distinct roles of Akt/PKB isoforms in cancer cell signaling. Studies have shown that Akt1 (PKBalpha) can attenuate breast cancer cell motility, whereas A...

LPLI inhibits apoptosis upstream of Bax translocation via a GSK-3beta-inactivation mechanism.

Low-power laser irradiation (LPLI), a non-damage physical therapy, which has been used clinically for decades of years, is shown to promote cell proliferation and prevent apoptosis. However, the underlying mechanisms that LPLI prevents cell apoptosis remain undefined. In this study, based on real-time single-cell analysis, we demonstrated for the first time that LPLI inhibits staurosporine (STS...

Investigating the role of signaling pathways and cancer stem cells in esophageal cancer with a therapeutic approach

Esophageal cancer (EC) is the sixth main cause of cancer death worldwide. Important genes associated with esophageal cancer include FOXO3, AKT, and GSK3β. Excessive FOXO3 expression inhibits the proliferation of cancer cells. The expression of AKT is involved in controlling cell growth in tumors. GSK3β activity is higher in cancer tissues. Given the effective role of cancer stem cells (CSCs) in...

PI3K/Akt inhibition and down-regulation of BCRP re-sensitize MCF7 breast cancer cell line to mitoxantrone chemotherapy

Objective(s):Multidrug resistance (MDR) of cancer cells is a major obstacle to successful chemotherapy. Overexpression of breast cancer resistance protein (BCRP) is one of the major causes of MDR. In addition, it has been shown that PI3K/Akt signaling pathway involves in drug resistance. Therefore, we evaluated the effects of novel approaches including siRNA directed against BCRP and targeted t...
